Ground Microbial Monitoring Strategy For Pharmaceutical Sterile Preparations in Cleanroom

In the production of sterile pharmaceutical preparations, ground contact disc testing in Class B areas is crucial for ensuring microbial safety.

Adhere to standards and meanings

USP1116 and GMP are the main basis. The former guides the testing operation, while the latter's microbial standards are applicable to the ground of B-level areas, strictly controlling the microbial limits in the production process. Key processes are carried out in the B-level area, with high requirements for sterility. Out of control ground microorganisms can easily contaminate drugs, and increasing monitoring frequency can prevent and control them in a timely manner.

Differences from C/D level areas

The production risk in C/D level areas is low, and monitoring requirements can be relaxed. The C-level area is used for material preparation, while the D-level area is responsible for auxiliary work such as external packaging. Enterprises can adjust the monitoring frequency of C/D level zones reasonably according to their own situation.

Determination of monitoring frequency in B-level area

There is no unified standard for monitoring frequency in B-level areas. Production of highly sensitive products or high-risk preparations requires increased monitoring frequency; Simple processes or low-risk formulations can reduce it. Referring to historical data, if pollution is stable, maintain the current frequency, and if there are large fluctuations, improve measures and increase frequency. When there are significant changes in personnel and equipment, the monitoring frequency should also be increased, otherwise it should be reduced.

Pharmaceutical companies should develop a scientific monitoring plan based on standards, combined with the characteristics of B-level zones and the situation of C/D-level zones, after comprehensive consideration, to ensure the quality and safety of drugs.
